Trump asks a Biden holdover to resign. They refuse. Here's what happens next:

Trump fires the holdover. The holdover refuses to acknowledge the firing, continues going to work and acquires legal counsel. All of their friends back up the holdover, flood social media with support, and threaten to quit in protest. Trump's cabinet encourages Trump to overlook it; it can't possibly be that bad to keep them employed. Stories about the event and of infighting in the Trump administration make the nightly news. The whole thing becomes a "scandal" and Trump backs down because it's becoming a colossal distraction. The holdover later becomes instrumental in pushing back Trump's America First agenda.

At the end of the day, Trump was waaaaay too worried about what people thought of him .. especially what the left thought of him.

Trump's resultant attempts at accommodation of his enemies may work in business, but it is a disastrous insecurity in politics where everyone gets ahead by trashing the person above them.
