The Constitution of the United States starts with the Preamble. We The People.....etc.
Then it goes on to lay out the three branches of the government: legislative, executive and judicial. It describes how they will work and who is eligible, how individuals are chosen etc.
The Bill of Rights then follow. These are the first ten AMENDMENTS of the Constitution. The first AMENDMENT is the freedom of speech, religion, right to peaceable assembly etc. The second AMENDMENT is the right to bear arms, etc.
Subsequent AMENDMENTS have been added to the first ten AMENDMENTS over the years.
We change the Constitution because we recognize it's not perfect. The Constitution itself provides a mechanism (actually three mechanisms), for its own change. Also, the 3/5th Amendment was struck down over a century ago. It was struck down fairly soon after the Civil War.
