No. The 25th Amendment, Section 4 says:

The "either" establishes the precedence:
(VP and (either Cabinet or Congress))
But, it only requires a majority if the President willingly goes along with it, or is unable to respond. If the President makes a counter-declaration that he is fit to serve, both the House and Senate must vote within 21 days, and a 2/3rd supermajority vote is required in each.

Cabinet, or the DOD, DOJ, or such. Executive departments are a pretty long list, but I doubt a letter from the principles from the Department of Agriculture is gonna carry much weight with invoking the 25th. It's the one part of the 25th that's never been invoked, for good reasons.
My worry is that the FBI is gonna go with her on it. They're an executive department, as defined by the Constitution - but she still needs the VP. And, all Trump has to do is write them an official 'I'm fine, you dumb, drunken cunts", and it's a non-issue.
Her going down that path, to me, is clear cut treason. I know it won't happen, but in a just world, Trump would let her try, and when she actually does it, haul her before a military tribunal under charges of treason and insurrection.

It's not about whether Pence will pull the trigger or not. They are going to try to send this through the Senate, which will delay ACB.
Without Pence, there is no path through the 25th Amendment. The VP has to agree to invoke it, period.
That's not how it works. It requires Pence to start the process before it ever even goes to the Senate.

My guess is that it gives congress the ability to pass a law saying another group can take the place of the cabinet in invoking the 25th amendment. I do not know of any restrictions on this group other than congress must vote on it into law.
I do not know of any such law that congress has passed, so it may be no such law exists right now and the only current way to trigger the 25th amendment is to vp + cabinet. But even if such a law has passed, the 25th amendment still requires the vp + the other group.
If it had to take a guess, assuming Pelosi was serious about invoking the 25th amendment and it wasn't just for PR, she is looking into if such a law was previously passed(again still requires vp + this other group), or writing a bill to create such a group that she know will try and trigger the 25h amendment.
Assuming Pelosi is writing a bill, it will have to pass through the republican senate before it can become law.. Even if a bill somehow passes the republican senate, GEOTUS will veto it. Even if SOMEHOW the veto gets overriden, vp would still have to agree with the group to trigger the 25th amendment.

So, VP & cabinet can invoke it. But if the president says there is no issue, "he shall resume the powers and duties of his office". Then the hurdle they have to clear is "Vice President and a majority of either the principal officers of the executive department", and then to congress, where "by two-thirds vote of both Houses that the President is unable to discharge the powers and duties of his office".
So impeachment and removal/conviction requires simple house majority and over 2/3 of senate. 25A requires VP, majority of cabinet, over 2/3 of house, and over 2/3 of senate.

The 25th amendment is designed so you can't remove a functional president. Its for brain dead ones or comatose ones, as even if they get the votes in the cabinet (which again will not happen) the president can return control to himself very easily.
There is already a mechanism for removing a president you don't like, its called impeachment, and she already tired that and it failed so badly they don't talk about it any more.
